NAP (Davunetide)

Intranasal

Overview

NAP, also known as davunetide, is an eight-amino acid peptide derived from activity-dependent neuroprotective protein (ADNP). It is thought to stabilize microtubules by interacting with tubulin and promoting cytoskeletal integrity, thereby protecting neurons from oxidative stress, excitotoxicity, and tauopathies.

Mechanism of Action

Intranasal delivery was pursued to achieve direct central nervous system access while minimizing systemic exposure..

Research Summary & Key Findings

Phase II trials in amnestic mild cognitive impairment and progressive supranuclear palsy were completed, but davunetide failed to meet primary cognitive or functional endpoints. A phase II/III trial in progressive supranuclear palsy was terminated early due to lack of efficacy, as reported in presentations at international conferences around 2012.

Clinical Status

Investigational

NAP (Davunetide) is in the investigational stage. While preclinical and early-phase data exist, it has not received regulatory approval for clinical use in the United States.
