Overview

Eltrombopag is a small-molecule, non-peptide thrombopoietin receptor agonist that binds to the transmembrane domain of the thrombopoietin receptor, distinct from the natural ligand binding site. While not itself a peptide, it mimics thrombopoietin signaling to stimulate megakaryocyte proliferation and platelet production.

Research Summary & Key Findings

Eltrombopag received FDA approval in 2008 for chronic immune thrombocytopenia, with subsequent expansions for severe aplastic anemia and chronic hepatitis C-associated thrombocytopenia. The RAISE trial (NEJM, 2010) demonstrated significant platelet count increases and reduced bleeding events in chronic immune thrombocytopenia patients refractory to other therapies.
