Klotho Peptide
Overview
Klotho is a transmembrane protein of approximately 1012 amino acids that exists in membrane-bound and secreted forms, with the soluble ectodomain functioning as a circulating factor. Klotho acts as a coreceptor for fibroblast growth factor 23 in phosphate metabolism and independently influences insulin signaling, oxidative stress, and cellular senescence pathways.
Mechanism of Action
Reduced klotho expression is associated with accelerated aging phenotypes in mice, while overexpression extends lifespan in rodent models. Therapeutic development focuses on peptide fragments or recombinant forms to replicate klotho's protective effects in age-related conditions..
Research Summary & Key Findings
Preclinical studies in rodents demonstrate that klotho administration improves cognition, attenuates kidney injury, and extends lifespan in some models. A small phase 1 trial evaluated recombinant klotho protein safety in healthy volunteers, confirming tolerability but not addressing longevity endpoints. No large-scale clinical trials have assessed klotho peptides for age-extension or healthspan outcomes in humans, leaving efficacy for longevity applications unproven.
Clinical Status
Klotho Peptide is in the research phase with limited clinical data in humans. Current evidence is primarily derived from preclinical (animal or in vitro) studies.
