Vantictumab

Intravenous

Overview

Vantictumab is a fully human monoclonal antibody that targets multiple Frizzled receptors involved in the Wnt signaling pathway. By blocking Frizzled receptors 1, 2, 5, 7, and 8, it inhibits Wnt-driven tumor growth and is designed to target cancer stem cells.

Mechanism of Action

The therapeutic rationale is based on dysregulated Wnt signaling in various solid tumors, particularly breast and pancreatic cancers. Inhibition of this pathway may reduce tumor recurrence and metastatic potential..

Research Summary & Key Findings

Phase I trials in solid tumors demonstrated dose-limiting bone toxicity including fractures, which restricted further development. A phase Ib study in metastatic pancreatic cancer combined with chemotherapy was terminated early due to safety concerns (2016). Despite promising preclinical activity against cancer stem cells, clinical development has been halted.

Clinical Status

In Clinical Trials
